The CD45 ptprc (Catalog #MBS666407) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The anti-human CD45 RO-Purified (with Antibiotics) reacts with Human, Primate, Monkey and may cross-react with other species as described in the data sheet. MyBioSource\'s CD45 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), FPS. Researchers should empirically determine the suitability of the CD45 ptprc for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The CD45 ptprc product has the following accession number(s) (GI #44242008) (NCBI Accession #AAS46946.1). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Testing Data
Information: Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. The CD45RO isoform contains none of the alternatively spliced exons and is present on most thymocytes, a subpopulation of resting (memory) T cells within both CD4 and CD8 subsets as well as on monocytes and granulocytes. CD45RO plays an essential role in lymphocyte activation. Antibody UCHL1 recognizes the CD45RO isoform of about 180 kd.
Performance: Five x 105 ficoll prepared human peripheral blood mononuclear cells were washed and pre incubated 5 minutes with 20 ul of 250ug/ml human IgG (to block nonspecific binding) after which they were incubated 45 minutes on ice with 80 ul of anti-CD45RO antibody at 10 ug/ml. Cells were washed twice and incubated with 2 degree reagent Goat anti-Mouse IgG/FITC, after which they were washed three times, fixed and analyzed by FACS. A net 25% sub population of the cells stained positive with a mean shift of 1.18 log10 fluorescent units when compared to a Mouse IgG2a negative control at a similar concentration. Production: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE with less than 1% Bovine Immunoglobulin.
Buffer: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 0.5 mg/ml Gentamicin Sulfate (as a preservative).
Group Header: anti-human CD45RO
Immunogen: Cultured human T cell line-CA-1. In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing CD45 are readily searchable from our website. Different antibodies against the same target such as CD45 may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
