Information: Human CD62P appears on the surface of activated endothelial cells or platelets and likely is involved with regulating the recruitment of leukocytes to areas of inflammation or infection (1, 2). Antibody G1 recognizes the protease sensitive lectin epitope of CD62P (1). Antibody G1 blocks the activated endothelium or platelet-neutrophil interaction (3, 4).
Performance: Platelet rich plasma (PRP) from heparinized blood was activated with 1 U/ml Thrombin in the presence of Citrate for 10 minutes. Ten ul of PRP per tube were fixed in 1% buffered formaldehyde, washed twice and pre incubated 5 minutes with 20 ul of human IgG at 250 ug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80 ul of anti-CD62P antibody at 10 ug/ml. Tubes were washed and incubated with 2 degree reagent Goat anti-Mouse IgG/FITC, after which they were washed again, re fixed and analyzed by FACS. A net 84% of the gated population stained positive with a mean shift of 1.83 log10 fluorescent units when compared to a Mouse IgG1 negative control. Production: Antibody was Protein A purified from (low FBS containing) tissue culture supernatant. Purity was >95% Immunoglobulin by SDS-PAGE with less than 1% Bovine Immunoglobulin.
