Flow Cytometry (FC/FACS)
Human CD66b (CEACAM8, CGMb, NCA-95, CD67) is a 95kD member of the Carcinoembryonic Antigen (CEA) group. It is expressed on granulocytes and functions as an adhesion molecule. Similar to the CD66 a, c, and d isoforms, engagement of CD66b on neutrophils causes increased adhesion to endothelial cells (2). Crosslinking CD66b on neutrophils with antibodies triggers IL-8 release (3).
Immunogen: Recombinant soluble human CD66b
Buffer: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 0.5 mg/ml Gentamicin Sulfate (as a preservative). Performance: Five x 105 Polymorphprep prepared human Granulocytes(PMN) per tube were pelleted and pre incubated 5 minutes with 20ul of human IgG at 300ug/ml (to reduce non specific binding) after which they were washed and incubated 45 minutes on ice with 80 ul of anti-CD66b antibody(ANC1D5) at 5 ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-Mouse IgG/FITC after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.28 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Binding was blocked when reagent was pre incubated with a 10-fold excess of recombinant CD66b. In general, we may offer more than one antibody to a given target to enable options for the researcher.
