Flow Cytometry (FC/FACS)
Human CD7 is one of the earliest T lineage markers and is found on most thymocytes and peripheral blood T cells. The function of CD7 is not yet fully known, although cross-linking of CD7 with antibodies induces a T cell transmembrane calcium flux and CD7 expression is induced by ionomycin. Antibody 3Ale recognizes a CD7 molecule of 40 kd. Antibody 3Ale induces T cell transmembrane calcium flux.
Immunogen: Human HSB2-T cells
Production: Protein A purified antibody from tissue culture supernatant was reacted with FITC. Unconjugated FITC was separated from antibody/FITC conjugate by desalting column. The antibody/FITC conjugate is at 0.5 mg/ml with a Fluorescein/Antibody molar ratio of 4.6. . Performance: Five x 105 ficoll prepared peripheral blood mononuclear cells (PBMC) were washed and incubated 45 minutes on ice with 80 ul of anti-CD7/FITC at a 1:50 dilution(10ug/ml). Cells were washed three times, fixed and analyzed by FACS. A 67% sub population of the cells stained positive with a mean shift of 0.95 log10 fluorescent units when compared to a Mouse IgG2b/FITC negative control (MBS666481) at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD7 antibody (MBS666367). Different antibodies against the same target such as CD7 may be optimized or tested for different applications and species.
