The CDK4 cdk4 (Catalog #MBS179152) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CDK4 Rabbit Monoclonal Antibody reacts with Human and may cross-react with other species as described in the data sheet. MyBioSource\'s CDK4 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC), Immunocytochemistry (ICC).WB: 1:1000-1:2000
IHC: 1:50-1:200
ICC/IF: 1:50-1:200
FC/FACS: 1:50. Researchers should empirically determine the suitability of the CDK4 cdk4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The CDK4 cdk4 product has the following accession number(s) (GI #4502735) (NCBI Accession #NP_000066.1) (Uniprot Accession #P11802). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the Anti-CDK4 Rabbit Monoclonal Antibody with the following immunoassay(s):
Western Blot (WB) (Western blot analysis of CDK4 expression in Hela cell lysate (MBS179152).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CDK4 monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CDK4 )
Ser/Thr-kinase component of cyclin D-CDK4 (DC) complexes that phosphorylate and inhibit members of the retinoblastoma (RB) protein family including RB1 and regulate the cell-cycle during G(1)/S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complexes and the subsequent transcription of E2F target genes which are responsible for the progression through the G(1) phase. Hypophosphorylates RB1 in early G(1) phase. Cyclin D-CDK4 complexes are major integrators of various mitogenenic and antimitogenic signals. Also phosphorylates SMAD3 in a cell-cycle-dependent manner and represses its transcriptional activity. Component of the ternary complex, cyclin D/CDK4/CDKN1B, required for nuclear translocation and activity of the cyclin D-CDK4 complex.
Immunogen: A synthesized peptide derived from human CDK4
Subcellular Localization: Cytoplasm. Nucleus. Membrane. Cytoplasmic when non-complexed. Forms a cyclin D-CDK4 complex in the cytoplasm as cells progress through G(1) phase. The complex accumulates on the nuclear membrane and enters the nucleus on transition from G(1) to S phase. Also present in nucleoli and heterochromatin lumps. Colocalizes with RB1 after release into the nucleus.
