Immunoprecipitation: 25ug/ml to immunoprecipitate rhCHIT1 from conditioned cell culture medium.
Neutralization: The antibody was preincubated with the enzyme at different molar ratios at RT for 30 min. The enzyme was then assayed with 4-MU-beta-D-N,N\',N\'\'-triacetylchitotriose. 50% of the enzymatic activity was inhibited by the antibody at approximately 56ng/mL (IC 50) under conditions in which the enzyme was present at 20ng/mL and the substrate concentration was 0.01mM. IC 50 was achieved at approximately 1:1 molar ratio of the antibody to the enzyme. Researchers should empirically determine the suitability of the Chitotriosidase n/a for an application not listed in the data sheet. Human CHIT-1 (chitotriosidase-1; also chitinase-1) is a 50kD, secreted glycoprotein that belongs to glycosylhydrolase family #18. It is synthesized by macrophages, and may serve to degrade chitin (poly-N-acetylglucosamine)-containing pathogens such as fungi and nematodes. Mature CHIT-1 is 435 amino acids (aa) in length, and contains a catalytic domain (aa #22-372) followed by a chitin-binding region (aa #373-466). Very high levels of CHIT1 are found in the plasma of patients with Gaucher\'s disease type I. Mature human CHIT-1 is 79% aa identical to mouse CHIT-1.
