The Hsc70 hspa8 (Catalog #MBS179527) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Hsc70 Rabbit Monoclonal Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. MyBioSource\'s Hsc70 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunoprecipitation (IP), Immunofluorescence (IF), Immunohistochemistry (IHC), Immunocytochemistry (ICC), Western Blot (WB).WB: 1:500-1:2000
IHC: 1:50-1:200
ICC/IF: 1:50-1:200
IP: 1:20
FC/FACS: 1:30. Researchers should empirically determine the suitability of the Hsc70 hspa8 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The Hsc70 hspa8 product has the following accession number(s) (GI #5729877) (NCBI Accession #NP_006588.1) (Uniprot Accession #P11142). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the Anti-Hsc70 Rabbit Monoclonal Antibody with the following immunoassay(s):
Western Blot (WB) (Western blot analysis of Calreticulin expression in (1) HeLa cell lysate; (2) C6 cell lysate (MBS179527).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HSPA8 monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for HSPA8 )
Acts as a repressor of transcriptional activation. Inhibits the transcriptional coactivator activity of CITED1 on Smad-mediated transcription. Chaperone. Component of the PRP19- CDC5L complex that forms an integral part of the spliceosome and is required for activating pre-mRNA splicing. May have a scaffolding role in the spliceosome assembly as it contacts all other components of the core complex. Binds bacterial lipopolysaccharide (LPS) et mediates LPS-induced inflammatory response, including TNF secretion by monocytes. Participates in the ER-associated degradation (ERAD) quality control pathway in conjunction with J domain-containing co-chaperones and the E3 ligase CHIP.
Immunogen: A synthesized peptide derived from human Hsc70
Subcellular Localization: Cytoplasm. Melanosome. Nucleus, nucleolus. Cell membrane. Localized in cytoplasmic mRNP granules containing untranslated mRNAs. Translocates rapidly from the cytoplasm to the nuclei, and especially to the nucleoli, upon heat shock.
Tissue Specificity: Ubiquitous.
