The Notch1 notch1 (Catalog #MBS802191) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Notch1 Antibody: ATTO 565 reacts with Mouse, Rat and may cross-react with other species as described in the data sheet. MyBioSource\'s Notch1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC).
1:1000 (WB). Researchers should empirically determine the suitability of the Notch1 notch1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.

The Notch1 notch1 product has the following accession number(s) (GI #224967065) (NCBI Accession #NP_032740.3) (Uniprot Accession #Q01705). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the Notch1 Antibody: ATTO 565 with the following immunoassay(s):
Western Blot (WB) (Western Blot analysis of Rat Brain Membrane showing detection of ~270 kDa Notch1 protein using Mouse Anti-Notch1 Monoclonal Antibody, Clone S253-32. Lane 1: MW Ladder. Lane 2: Rat Brain Membrane (10 ug). Load: 10 ug. Block: 5% milk. Primary Antibody: Mouse Anti-Notch1 Monoclonal Antibody at 1:1000 for 1 hour at RT. Secondary Antibody: Goat Anti-Mouse IgG: HRP at 1:200 for 1 hour at RT. Color Development: TMB solution for 10 min at RT. Predicted/Observed Size: ~270 kDa.)
Western Blot (WB) Notch1.

Background Info: Detects >270kDa, ~120kDa and small fragments due to proteolysis. Does not cross-react with Notch2 or Notch3.

Scientific Background: Notch is synthesized in the endoplasmic reticulum as an inactive form which is proteolytically cleaved by a furin-like convertase (S1 cleavage) in the trans-golgi network before it reaches the plasma membrane to yield an active, ligand-accessible form. Cleavage results in a C-terminal fragment N(TM) and a N-terminal fragment N(EC). Following ligand binding, it is cleaved (S2 cleavage) by TNF-alpha converting enzyme (TACE) to yield a membrane-associated intermediate fragment called Notch extracellular truncation (NEXT). This fragment is then cleaved by presenilin-dependent gamma-secretase (S3 cleavage) to release the intracellular domain (NICD) from the membrane.

Conjugate: ATTO 565. Storage Buffer: PBS pH7.4, 50% glycerol, 0.09% sodium azide. In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing Notch1 are readily searchable from our website. Different antibodies against the same target such as Notch1 may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
