The PD-L1 cd274 (Catalog #MBS179113) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PD-L1 (CD274) Rabbit Monoclonal Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. MyBioSource\'s PD-L1 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC), Western Blot (WB).WB: 1:100-1:500
IHC: 1:50-1:100. Researchers should empirically determine the suitability of the PD-L1 cd274 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The PD-L1 cd274 product has the following accession number(s) (GI #390979639) (NCBI Accession #NP_001254635.1) (Uniprot Accession #Q9NZQ7). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the Anti-PD-L1 (CD274) Rabbit Monoclonal Antibody with the following immunoassay(s):
Western Blot (WB) (Western blot analysis of PD-L1 (CD274) expression in Ramos cell lysate (MBS179113).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD274 monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD274 )
Immunohistochemistry (IHC) (Immunohistochemical analysis of paraffin-embedded human placenta, using PD-L1 (CD274) Antibody(MBS179113)
PD-L1 (CD274) was detected in paraffin-embedded tissue section. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti- PD-L1 (CD274) Antibody (MBS179113) overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.)
Immunocytochemistry (ICC) (immunocytochemical analysis of SCLC cell, using PD-L1 (CD274) Antibody(MBS179113)
PD-L1 (CD274) was detected in SCLC cell section. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The cell section was incubated with 1ug/ml rabbit anti- PD-L1 (CD274) Antibody (MBS179113)overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.)
Involved in the costimulatory signal, essential for T- cell proliferation and production of IL10 and IFNG, in an IL2- dependent and a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation and cytokine production.
