Note: Immunofluorescent assays were performed using methanol-fixed HCT8/E8 and paraformaldehyde-fixed HaCat cells. For optimal IF results, methanol and paraformaldehyde- fixed cells should be permeabilized with 0.2% Triton X-100 for 5 and 15 minutes, respectively, prior to incubation with primary antibody. Researchers should empirically determine the suitability of the PKP3 pkp3 for an application not listed in the data sheet. Plakophilin-3 is a novel 87kD Armadillo-like (Arm) protein, which is characterized by a series of repeats of 42-45 a.a., originally described in the Drosophila protein Armadillo. Plakophilin-3 is a member of the p120ctn/plakophilin subfamily, which includes plakophilin-1, plakophilin-2, the ARVCF gene product and d-catenin. This protein is similar to plakophilin-1 and plakophilin-2. Other Arm proteins include b-catenin and a-catenin. All these proteins are part of the cell-cell adhesion complex. On Western Blot analysis, plakophilin-3 was expressed in epithelial cell lines but not in foreskin fibroblasts and sarcomaderived cell lines. By Immunofluorescence, plakophilin-3 was colocalized with desmoglein in desmosomes of epithelial cells.
